Description: Hemiasterlin is a linear tripeptide isolated from the sponge Hemiasterella minor in 1994. It contains two unique amino acids and exhibited strong cytotoxic activity on the P388 leukaemia cell line. Hemiasterlin is a potent inhibitor of cell growth that, like vinblastine, depolymerizes microtubules and arrests cells in the G2-M phase of the cell cycle. However, like other peptide-like antimicrotubule agents, such as dolastatin-10 and cryptophycin-1, hemiasterlin noncompetitively inhibits the binding of vinblastine to tubulin. Because hemiasterlin competitively inhibits the binding of dolastatin-10 to tubulin, it has been proposed that these molecules bind to the Vinca-peptide-binding site of tubulin.
